Biotin, Collagen Peptide and Keratin Tablets

Three actives sit in this tablet and they come from three unrelated places, which is why the specification treats them separately rather than as one beauty complex. D-biotin (CAS 58-85-5, EC 200-399-3, C10H16N2O3S, molecular weight 244.31, PubChem CID 171548) is vitamin B7, a bicyclic molecule of a ureido ring fused to a tetrahydrothiophene ring, bought on the USP assay of 97.5 to 102.0 percent on the dried basis and made by chemical synthesis, so the biotin input itself carries no animal origin. Hydrolysed collagen peptides are enzymatic hydrolysates of type I and III collagen from bovine hide and bone, or type I collagen from fish skin and scale, bought on protein content - typically not less than 90 percent on the dried basis - with average peptide molecular weight in the 2,000 to 5,000 Da band; the protein keeps the Gly-X-Y repeat with glycine at roughly a third of all residues, and hydroxyproline is the marker actually assayed. Hydrolysed keratin is the third and least standardised of the three, recovered from sheep wool (Ovis aries) or poultry feather (Gallus gallus) by sulphitolysis, oxidative or enzymatic routes, and it is bought for its sulphur amino acids, cysteine being reported at roughly 10 to 17 percent of wool keratin's amino acid content, far above ordinary dietary protein. The format is where most briefs for this product go wrong. Biotin is dosed in micrograms and disappears into a tablet without difficulty, but collagen and keratin are dosed in hundreds of milligrams to grams and are bulky, hygroscopic and poorly compressible. A tablet of workable size carries a fraction of what a powder sachet carries, so the 5 to 10 g collagen servings seen on powder packs are simply not a tablet proposition; an honest tablet build declares collagen and keratin in the low hundreds of milligrams and says so on the panel. Fixing the per-tablet load before tooling matters, because it drives tablet weight, punch size, blister cavity and the number of tablets in the daily dose. The excipient set is conventional - microcrystalline cellulose, dicalcium phosphate, croscarmellose sodium, magnesium stearate and colloidal silica, with an HPMC film coat - and because both protein hydrolysates pick up moisture, granulation, compression and packing run under controlled humidity with alu-alu blister as the preferred barrier. Origin is the declaration to read first. Collagen and keratin are both animal proteins, so no build of this tablet is vegan or vegetarian whatever the coating is made of, and the species has to be pinned down at order stage. Fish collagen is a declarable allergen under Annex II of Regulation (EU) 1169/2011 and must appear on the label; bovine collagen is not a listed allergen but raises TSE documentation and religious-certification questions; wool keratin brings its own ovine declaration. Beyond those inputs the allergen picture is clean, with no milk, egg, soy, peanut, tree nut, crustacean or gluten material used. On wording, only biotin carries an authorised EU health claim relevant here: Regulation (EU) 432/2012 permits the statement that biotin contributes to the maintenance of normal hair where the product qualifies as a source of biotin against the 50 µg nutrient reference value in Annex XIII of Regulation (EU) 1169/2011, while collagen and keratin have no authorised EU claim and must be described compositionally. One further point belongs in the consumer leaflet rather than the trade file - US FDA safety communications issued in 2017 and updated in November 2019 warn that supplemental biotin can interfere with immunoassay laboratory tests, including falsely low troponin results, and many brands now carry a line advising users to tell their clinician.
