Beta-nicotinamide mononucleotide (NMN) is the 5'-phosphate ester of nicotinamide riboside: a nucleotide assembled from nicotinamide, D-ribose and a single phosphate group, molecular formula C11H15N2O8P, molecular weight 334.22, CAS 1094-61-7. The IUPAC name is 3-carbamoyl-1-(5-O-phosphono-beta-D-ribofuranosyl)pyridin-1-ium. The trade article is the beta anomer, the form the NAD biosynthetic enzymes recognise. Metabolically NMN sits one enzymatic step from nicotinamide adenine dinucleotide: nicotinamide phosphoribosyltransferase forms NMN from nicotinamide, and NMN adenylyltransferase converts NMN to NAD. It is a distinct article from nicotinamide riboside, which carries no phosphate group, and from NAD itself, which is a dinucleotide - the three are not interchangeable on a weight-for-weight basis and each carries its own specification and regulatory position. Bulk NMN reaches the market by two routes: chemical phosphorylation of nicotinamide riboside, which depends on phosphoryl chloride and tight temperature control, or an enzymatic route in which an engineered nicotinamide riboside kinase phosphorylates NR with an ATP regeneration system. Both give an entirely non-animal, non-dairy article. The powder is white to off-white and freely water soluble - Sigma-Aldrich reports roughly 50 mg/mL to a clear, colourless to faintly yellow solution - and it is hygroscopic, so it ships under nitrogen in aluminium foil with desiccant and is best held cool and dry. Formulators taking it into effervescents, sublingual tablets or ready-to-mix sachets should control water activity in the blend and in the finished pack, and avoid prolonged heat during granulation. Regulatory position is unusually mobile for this ingredient and buyers should read it market by market. In the United States, FDA excluded NMN from the dietary supplement definition in November 2022 under the drug preclusion clause, then reversed that position in letters dated 29 September 2025 and reinstated NDI acknowledgements in December 2025. In the EU, NMN is not yet on the Union list of novel foods; EFSA adopted a positive safety opinion on beta-NMN on 11 May 2026 against an application filed on 2 August 2023, and the Commission implementing act is still pending. Japan added NMN to its food raw material list in March 2020. China's National Health Commission has not approved NMN as a food additive or new food raw material. In India, FSSAI stated in a notice dated 24 July 2026 that NMN is an unapproved ingredient for supplement use and that the non-specified food and food ingredients route must be followed for domestic sale.
